后来他到了丹佛，在那里，他在里吉斯学院学习医学，但因为携带大麻被捕，中途退学。随后，他和伙伴丹·艾克洛德，吉尔达·拉德纳以及约翰·贝鲁希一起加入《全国讽刺电台一小时》(National Lampoon Radio Hour)栏目。后来，其他三名伙伴成为《周六夜现场秀》(Saturday Night Live)节目创办人，他加入了《霍华德·考塞尔周六夜现场秀》(Saturday Night Live with Howard Cosell)节目。《霍华德·-考塞尔周六夜现场秀》宣布失败后，他才有机会加盟《周六夜现场秀》，并那里很快成名，为以后电影界发展打下基础。

1979年，比尔·默瑞出演第一部影片《肉丸》(Meatballs)，从此一发不可收拾。随着1982年的《杜丝先生》和1984年的《捉鬼敢死队》的卖座成功，比尔·默瑞一跃成为80年代炙手可热的喜剧明星。1990年，比尔·默瑞开始尝试执导，影片《两男一女三逃犯》的成绩尚可。而后几年的喜剧作品，维持了其一贯的个人风格。在影片《霹雳天使》中也有不错的表现。2003年，比尔·默瑞主演了女导演索菲亚-科波拉(Sofia Coppola )小成本电影《迷失东京》(Lost in Translation)，并凭此片入围第76届奥斯卡金像奖最佳男主角。
